Taking the train is the first option. There is a line connecting Hong Kong to Guangzhou. It takes about two hours and there are several trains heading back and forth each day. You just have to make advance reservations to avoid delays and other problems. This is not the most comfortable choice though. This train system is also not the best you can find out there. If you are on a tight budget, this could work.
If you want to get there at the fastest possible time, there are a few scheduled flights each day. The entire flight takes about an hour. If you are willing to spend more, this is the best choice for you. Just remember than you still have to head to the airport in Hong Kong, check your luggage in and do the same thing upon arrival in Guangzhou. It seems like too much of an effort for a really short trip. This is on top of the amount that you are spending for the ticket.
The best option
Taking a private transfer Hong Kong to Guangzhou and vice versa is the best choice. It is totally comfortable and relaxing. You just have to make advance reservations, indicate the place where you will be picked up and decide on the date and time. You also need not worry about your local transportation in Guangzhou. You already have a car that will take you to different places. You just have to relax. The Toyota Alphard and Mercedes Benz S350 are great choices to help you enjoy your trip even more.
The road to Guangzhou from Hong Kong is not necessarily the best. You also have to go through immigration checkpoints. The face that you are in a comfortable vehicle like the Mercedes Benz W221 or Benz E-Class (Golden) will make the travel better for you. The cost might be quite high since you are renting the entire vehicle, but it is relaxing. You have nothing to think about anymore. You will also save more time especially if you intend on visiting different places.
